Most of the guesses about the mom's illness, are that it's Tuberculosis, which would fit with the 1920's / 30's esthetics in the film. Also, Hayao and his brothers saw their mother go through long treatment for Spinal TB when he was a child, she was treated over 9 years, including multiple stays at the hospital, so many see this portion as probably being at least, partially, autobiographical. Given his personal history, I think it's safe to assume TB; the only other major illness that I can think of, that fits the approximate era of the film, is Polio.

Good news and bad news: there was a short sequel! The bad news: it can only be seen in the theater in the Ghibli museum in Japan. You can find still images though, and they're beautiful. The film features Mei and a KITTENBUS as well as more totoros. It's really tough not to cry for the children in the third act. It adds a lot of weight to the story. We have to assume from the end credits that mom gets better.
