I don't know why some people complain about this video. I find it is amazing and very helpful. I'm undergraduate and my research project used Y2H for the experiment. After watching this video I understand the concept of this Y2H completely. I like how you explained that the Y2H is like making a "check point" by using the yeast organism as a demonstration. I usually don't write a comment but you should give a thump up bro! For those who don't understand your language and accent, I find it's helpful to turn on the CC, pause the video and write down what he said. Some science concept are really complicated and you need to paused and break down each points. There are a lot of information in this video so it's not just really his fault about explaining things. It's about your efforts trying to learn it from it as well. Anyway, your channel is truly amazing bro! :))

How would you know which bait and prey proteins to start with? For example if you had the blue crescent one as bait and another prey protein that doesn't fit, they won't bind to the transcriptional activator binding site, therefore no transcription nor translation. But does that then mean you need to plate a Petri dish for every possible protein in the cell? e.g. plate #1 has bait as blue crescent and prey as a purple bulb, plate #2 has bait as blue crescent and prey as a red bulb etc etc

Great video! I understood that this method is usually done in a matrix of thousands of proteins X thousands of proteins. After we get the interaction pattern, how can we trace which are the exact interacting proteins?

Does this always work? How can you tell if the orientation of the bait and prey attached to the BD and AD will make it start translation? If you attach it "wrong", wouldn't a dimer be so misformed that it wouldn't translate regardless of a successful bind?

What is stopping the transactivating domain protein (prey) binding the bait if the target protein and partner do not bind? There must be a system in place to stop all the false positive readings, can someone explain that to me?

Shomu, I am sorry to say that both your explanation of yeast two-hybrid (y2h) system and your language is very poor. Initially, one has to begin describing in a very simple way explaining what it is, how and why it was developed, elucidate the PRINCIPLE of y2h system elaborating on each component of the assay separately, explain the methodology of reporter assay and later describe how it is useful and different from other assays etc. I have used this assay many times in my work and I did not find your explanation coherent, logically framed step- by- step that could be useful for students who do not even know what y2H is!

+Reginald Finley yes, just like I did when I learned English because it wasn't my first language. The reality is that a lot of people have trouble understanding foreign accents. Fixing your accent to be more articulative is just more professional. Especially when your job involves lecturing. If you have ever been to a few university lectures you know what I'm talking about. And yes, maybe it's not completely PC to say this but it's the truth.
