Your videos are a treasure! One year ago I watched your DIY autoclave video and next week I’ll be starting a job in plant genomics. On paper, I’m completely unqualified. This will be extremely useful for growing cactus from seed. When we grow cactus from seed, we typically grow it under cover for weeks or months or even years, so the seed doesn’t dry out and die or get stunted. Cactus seedlings have minuscule root systems and it takes ages for them to develop. Normally we sterilize the soil in the microwave and then grow it in some kind of plastic container with a lid. I’ve had a lot of failures with this method due to mold and algae finding their way inside and either suffocating the seedling or causing damping off. Next winter I wanna abandon my caveman ways and do it the proper scientific way.

Since cactus seeds are soooo tiny, you could also use a micropipette with sterile tips to pick up the seeds in the solution and place them directly onto the media. I am planning to try that soon with some gibbaeum seeds

When I first learned how to do this I had the biggest nerdiest smile on my face thinking about how I could use this method. I don’t have the equipment since it was the companies equipment and I wasn’t really looking because of how expensive it typically is for their methods. Though seeing your video makes me want to get back into it and buy the stuff for my own garden.

I came here to learn about plant tissue culture techniques, and stayed not only for the marvellous content, but because you are the sexiest, funniest nerd I have ever seen! I used to love watching the Big Bang Theory, and I have to say, if you took the three leading ladies ie Penny, Bernadette and Amy aka Melissa Rauch, Kaley Cuoco & Mayim Bailik and somehow amalgamated them together, you would get YOU! Beauty, pazazz and brains. I know these days that you aren’t supposed to say things about peoples looks etc, but I just had to! Also, your humour is off the charts dry and funny as heck. Yes 100%. But since the seeds are so small I would use a sterile micropipette to transfer them instead of forceps.

I have to say this is the video that woke me up to this❤. I'd seen so many tissue culture videos on FUNGI, not with plants, at least by how I was searching. My first experience with it was really at Disney World, the place known as pleasure island, had some really cool stores and they sold sequoia trees in testtubes. and of course they were adorably tiny in there and I wanted to buy one, but I was afraid I'd kill it as I didn't know sooo much at 15 years. I think you're channel is a gem and your vocal delivery has a jovial personality that brings an inner child to life within me as I watch and learn from you. May you in all facets of your life know true, blessed joy.❤❤

I've always used polypropylene plastic bags for sterilizing tweezers and other hardware. And you can re-use them for 10's or even 100's of times.

Back when I worked with mammalian cells we would give them a bit of penicillin/streptomycin to reduce the risk of contamination. It's probably poor practice due to the whole antimicrobial resistance issue we've got going on, but it allows the students to not contaminate all of their samples.

I also like to make 100s of plants from one plant but I take the cheaper longer route and do traditional cloning with plant cuttings.

Interesting to know how easy this process is once you have all the equipment and knowledge. Thanks for making this video to educate us. However, I don’t mind waiting a year to collect seeds from the best plants as that’s what I do every year anyway.

amazing video!! one question, 5ml of h2o2 and 45ml of water gives 0,3% of final solution, that is not very weak to sterilize? usualy I like to use 1% solution

PH meters are ALL garbage and failure is a question of when, not if. Use PH test strips instead.

Vanilla is really easy to clone in TC. To get a specimen sized plant to produce a seed pod is another story lol. Saffron also yes, but TC won't make it cheaper because the problem is each plant yields very little. Truffles are fungus so I know nothing about them...

I’ve had this thought in my head a few days… if you add two types of the same plant like say Thai constellation and a albo monstera could you cause a hybrid in tc? Maybe if the tissues are “grafted” to each other might help get some cells to swap. Just a thought, could be a cool experiment.

That’s exactly how it works, it’s actually a breeding technique that already exists. What u described creates a chimeric cross. Although not a hybrid because the plants u described are in the same species. U could take it a step further by a another method known as “Cellular Fusion” by coating the cells with polyethylene glycol, which dissolves away the membrane, letting the cells merge, replicating natural hybridization. Nowadays u don’t need to wait till maturity.

Naturally cells will not merge unless the membrane is penetrated… if u remember in school… reproductive cells has the ability to penetrate the other team thus allowing fusion, while with vegetative cells that layer has to be removed using various ways.

@@mil0thebird possible, u can even do it without TC. Find a Thai constellation monstera stem with a bud cut off the stem just right in the spot it's growing a bud clean and straight with precise tools, try your best to make an exact half cut of the bud-junction and do the same to the pink princess. Connect the top half with the bottom half. And apply keiki paste on the bud eye. Now take good care. pruning off other potential growth points may lessen the wait although not nessasary.

I find autoclaving typically crushes polypropylene containers, do you find that not sealing them helps?

The larger ones sometimes do get crushed if you stack them. If you have a laminar flow hood, you can pour the media under the hood. The small ones I use from lollicup also do not crush in the autoclave, even when stacked. Hopefully that helps!

I bought some from Phytotech when I was first getting started. Not sure if they still sell it though. Once you get the hang of it, the media is very easy to make.

I would think this would work for any seed, with some modifications. Note that some seeds require stratification to germinate - you would cold treat the seed and then follow the procedure.
