Amazing video. It’s definitely gonna be useful. I actually managed to find a source for the chemicals and plant hormones needed for TC. Seed banks here in Europe release their catalogs of seeds around fall, early winter, so it’s a project for later in the year. I’m mostly interested in germinating seeds using TC methods. Cactus seeds are super fussy when they’re young and need constant humidity. I get so many losses to algae and fungi.

Kind of related to TC is a more low tech approach. In Takeaway tec or coke bottle tec you place the cactus seeds in a closed container with soil, preferably sterilised to some degree. The small cacti can handle the high humidity just fine, I've had plenty that shared the soil with algae. For propagating cactus plants grafting is great. All cacti can be grafted to each other. It's great fun and really rewarding. Combining both can yield big cacti in a relatively short time.

For comercial growers TC is a big thing in the germination of many very small and high cost seeds, such as orchids, maybe you could try to germinate store-bought vainilla seeds or something like that, just for fun

The way you draw the seeds up into the pipette, I do the same thing with fungal spores for germination on PDA media. It's wild how similar the two practices are.

Quick question; when growing succulent seeds in vitro, should they be put on a germination mat? When germinating seeds the conventional way, a heat mat is a must for succulent seeds. The seeds I’ve tried to start in vitro rarely germinate. Let me know what you recommend. Thanks!!

No, I think that would melt the media. I recently had issues getting echeveria seeds to germinate though, so maybe temp was the issue.

Hey, for the bleach rinse step, do you reuse your bleach solution for the 3 rinses? Do you need new water for the water rinse step, or can you reuse the water for the rinse? What temp do you bring your medium recipe to, to be sure the agar dissolves? thanks for all your awesome videos!

No, I wouldn't reuse the water. I just try to have enough on hand to rinse the plants enough times. And I don't use heat when I make the media most of the time, I just throw the whole media bottle in the pressure cooker / autoclave

Should the autoclave be under the flow hood, along with the shaker? I’ve gotten contamination and I wasn’t sure if the open air transfer was an issue, even if the jar is sealed with a lid. (Not plastic wrapped though) I’ve googled how to asses contamination because I’m curious if it’s from my explant-succulent, like a fungus or bacteria. It had some tiny black specks on it where it looks like necrosis of the plants epidermal layer. My microbiology class was a while ago, but I know there’s a series of tests you can use to narrow down the issue. I’ve been looking on reasearchgate with not much luck. I’d love a video on how to take the contamination and read the results under a microscope, to see if my plant is sick or it’s my “dirty house air”, lol. How about using a sterilized dilute solution of sodium thiosulfate as a rinse after the bleach bath? That should neutralize the chlorine/stop bleach activity and minimize the number of rinsing needed. So something like bleach bath, then sodium thiosulfate rinse, and then a water rinse.

The biotilt is interesting to me, but the entire system is so basic and unpolished as to make me think I should just build it myself (that is, assuming it's pretty expensive to buy from PCT). It also looks like it needs a much better solution for locking in the biocouplers. How do you examine the state of a coupler in the middle of a rack, surrounded by other couplers? Lean into the biotilt and peak into the jar from above? I wonder if early detection of issues would be made more difficult that it should be. Perhaps I'm misunderstanding how it works too, but it looks like you need to take out an entire row of couplers at a time to inspect them individually. In any case, I'm looking forward to your video about it!

Thanks for your videos. I'm going to give tissue culture a try for the first time. Just a heads up, "sterilize" is different than "sanitize". Sterilize uses heat, sanitize uses chemicals. So technically you're sanitizing the nodes when you put them in the bleach solution, not sterilizing them. But whatever no big deal.
