a vector is represented by a single column matrix 3x1 now, think this way: a body in space will be represented by its position and orientation,. position vector is a single vector(3x1) whereas orientation vector is a combination of three vectors(3x3), thus the representation of a body in space is a combination of four(4) vectors in total. hence it will be a 3x4 matrix

we are not multiping them we are just writing them together. so, 3X1 | 3X4 gives 3X4 matrix

It is a 3x3 matrix, as uXb, uYb, uZb are all 3x1 vectors. Expand them, and you will get a 3x3 matrix.
