How can a single injection have such lasting result? Why isn't the siRNA consumed and the cell 'functions' returned to normal?

The LINE-1 pathway in which mRNA can be reversed transcribed into DNA copies which are inserted into the genes of that cell is a probable explanation. That way, the cells can keep generating new mRNA for new protein.
