Well the thing is, at the beginning it's less hatred and more greed plus jealousy. He hated that Jonathan got a much better childhood than him and wants the entire Joestar inheritance to himself. That's why he wanted to mentally break Jonathan. It was so that when he killed the Dad, he could just take everything under the Joestar name without Jonathan having the spirit to contest anything. But Dio also has an extremely fragile ego, and hates getting hurt or embarrassed, having had more than enough of that as a child. So when Jonathan beats him up and subsequently brings him back to that helplessness and despair he felt as a child, even if only for a second, a deep seated feeling of hatred began to form. At the end he didn't even really hate the bloodline that much and it was Joseph and Jotaro who decided to go after him while he was minding his own business. Of course, they had no choice in the matter but to DIO, it's the same either way. This damn family that put you in time out for 100 years is now coming for your life. I wanna see the person that wouldn't be angry. The only part that I really see as petty hatred is when he was dead but his influence was still ruining the lives of the Joestar family and co even decades later. That is true commitment to the bit even if not done by way of his will.

It had something to do with the Part's main villain in an by-extension sense, but really (without getting into much details) Part 7 Jojo's death was more to do with a Monkey's Paw kind of mystical trade-off in the form of an inherited disease.
