I am 40 years old, with low back pain and left knee pain since my early thirties, it was tolerable most of the time and sometimes (twice a week) I needed NSAIDs. I started at the beginning of last year with 3mg which I escalated weekly up to 5mg per week and by the third week I was pain free, the only side effect was insomnia on the days of use. In total, 9 months of continuous use, and now in my third month of rest, the pain has returned but more attenuated. I plan to restart next month. Thank you for all of your work

My wife and I've been taking 6mg/week for 1.5 years, with a total of 4 months off (6 on, 2 off, 6 on, 2 off). Her lifelong arthritis is significantly better and her overall health has improved. My shoulder injury pain (from renovation) that would not go away is almost cured but teeth health questionable. Got the shingles vaccine recently, hoping Rapamycin will further improve efficacy. We'll probably continue as long as we can afford and until proven otherwise.

Thanks for your work, and objectivity even in this compelling N=1 story. I appreciate your continual comments about the potential for placebo, and sharing the rationale for your trial. I have been cycling 5-6 mg/week on and off for a couple of years. I had experienced a similar shoulder challenge when lifting and would moderate the weight on bench press and shoulder press. The pain would occasionally make it impossible for me to lift heavy for weeks, but thankfully this has not been an issue for a couple of years. Is rapa the causal mechanism for relief from the pain? Maybe, just have to wait for some human trials to get more evidence.

Hey Matt. I had frozen shoulder in both of them some years ago. And the doctor told me that there wasn't a lot I could do. It went on for more than a year. Then I decided to fast for 5 days (water only) to lose some kilos. After the fasting period my frozen shoulders were healed. I did some research and found out that during fasting hydroxy-butyrate is produced which has an anti-inflammatory effect. Now I am starting to think that it might have been because of a downregulation of mTOR.

Thanks, Matt. I just started rapamycin. I'm in my third week (5 mg per week). I started very late in life (74 years). I wish I had started earlier. I'm battling plantar faciitus in my left foot. It's been getting better since before the rapamycin, but I'm hoping it helps me get beyond it.

In an upcoming video maybe you could address the following; In the NIH ITP studies, were there a difference in cancer mortality rates between the controls and Rapamycin cohorts?

Good question. My recollection of that data is that the tumor burden at time of death was comparable between rapamycin treated and untreated but, of course, the time of death was significantly later in the treated animals. One interpretation of this is that rapamycin *delayed* cancer progression but did not prevent it, at least at the doses used in the ITP studies. One minor point that's also worth noting is that we don't really know what the cancer mortality rate is in either case, because most laboratory mice are euthanized before they die. Sometimes this is because of visible tumors, but often it's for more nebulous reasons like unexplained weight loss above a pre-specified threshold. That could be caused by cancer but could also obviously be caused by other underlying pathologies.

@@houndofzoltan Yes, and no. The ITP mice are a mixed background, so it's probably not due to their breeding. More likely, it's simply that mice when allowed to reach old age (which they don't in the wild, due to predation) are more prone to cancer than humans *because* there has been little selection to prevent cancer (precisely because they never reach 3 years of age in the wild). Regardless, rapamycin delays or reverses age-related functional declines and pathology in every tissue and organ where it has been studied, so it's clear the effects of rapamycin go far beyond simply impacting cancer.

I’ve been taking 9 mg 1x per week on a 8 week on 5 week off cycle for about 8 months. I haven’t had a single side effect. I’m actually considering upping it to 10mg on the same schedule. The dose given to dogs in your study at 1mg per kg would equate to 10mg for me.

We've tested three doses in our dog studies so far: 0.1 mg/kg given 3x per week, 0.05 mg/kg given 3x per week, and 0.15 mg/kg given once per week. It's tough to do a true dose equivalency, but a rule of thumb would be to divide the dog dose by 2: www.ncbi.nlm.nih.gov/pmc/articles/PMC4804402/ - MK

What is the purpose of cycling? I’ve been taking Rapa for three years without any breaks. The only issue I had was a short period of mouth sores. On a positive note, I’ve had a slight decrease in appetite.

I understand your reasoning why rapamycin caused the relief, but ... ... the specialist told you this goes away by itself PS - I do find you agreeably skeptical pretty often, and am listening to you in preference of the more cheerleading crowd. But I may be more skeptical than you in this area.
