In answering the question about dosing of rapamycin, my understanding had been that you advised taking it for 10 weeks, and then taking a break for 6 months or more. Is that still the case?

Matt, In my opinion, this has been the most interesting interview you have done so far, Professor Aduti Gurkar was effusive and graceful, with no ego, just putting forward excellent information. For me, as a layman, it was very timely as I have been researching all the science on age related autoimmune illnesses and immunosenescence, becaused in September last year at the age of 62, I was diagnosed with IPF an autoimmune type, called nonspecific interstitial pneumonitis (NSIP) with all sorts of secondary autoimmune manifestation like sjögren's syndrome, antisynthetase syndrome, myositis, polymyositis. The IPF diagnosis is 3 to 5 years life expectancy. It's as though my body is attacking me from the inside, but to look at me you wouldn't know it, I look younger than my age, and within 5kg of my body weight when I was 25 and do HIIT exercises, and have always eaten a balanced healthy diet, nothing processed only fresh vegetables and meat, never take away food, I was a fitness fanatic in my youth doing martial arts. What I do know is that we all age at different rates, and for some, we have to fight our genetics, if we're to extend our life span. It's a role of the dice at birth. I am now following research done by the Paul Nobel Lab at Cedars Sinai Hospital in LA, into pulmonary fibrosis where trials on mice and in vitro cells resolved the fibrosis due to finding that the ZIP8 (SLC39A8) zinc transporter not functional, stopping zinc getting to the type 2 alveolar cells to repair the fibrosis. The challenge is to reactivate the zinc pathway to the lung. They found that combining the zinc supplementation with NMN and Resveratrol significantly increases the zinc getting to the lungs cells and the resolution of the Fibrosis. The research paper is available online. This has created my obsession to understand and dig deeper into age related illnesses and what is happening to the ageing immune system and increase in autoimmune diseases as we age, I learnt about senescence cells and Senolytic supplements, and given the IPF life expectancy, I am willing to hope against hope that the Noble Lab treatment is worth a shot in the dark, I have also added Lipsomal Quercetin, Lipsomal Fisetin, Pterostilbene and EPA DHA Omega-3 to my treatment. The dosage I'm taking is very high, double the recommendation, including the zinc. I feel better than I did when first diagnosed and my pulse ox reading is now better, a solid 97 or 98, last year I was lucky to get 93 or 94, my 6 minute walk tests are about 700 meters, any faster and I would be running. The confirmation if this treatment works will be in March / April next year when I have my second lung CT scan, to see if the fibrosis is growing or receding. Could you PLEASE interview leading research scientists working on autoimmune illnesses and its relationship to the ageing Immune system, I think there must be a link between autoimmune illnesses and the lingering senescence cells causing autoimmune illnesses, which are common as we age and affects our health-span. Please have Aditi Gurkha back and explore this fascinating subject more. It would be great if you interviewed Professor Paul Noble of the Noble Lab at Cedars Sinai Hospital, he is a pulmonologist doing research over the last 25 years, he does do interviews and the human clinical trials of the zinc, resveratrol and NMN has recently begun. Thank you, Matt, and keep up the good work, Enrico, Melbourne Australia. Cheers.

Really?! You sound equally stupid calling a banana the same as a Snicker's bar. Bananas not a healthy food? Fiber, potassium, etc. Use a green (underripe) banana that has higher resistant starch content or frozen banana to prevent ripening and promote resistant starch, if you are someone that obesses of glycemic response. You go on to name groups that have metabolic imbalance or pre-diabetes, those are groups that should be taking steps beyond worrying about bananas. --- This video just stands as an example why a research scientist shouldn't be mouthing off about nutrition on social media. Not your wheelhouse.

Dosage Even if we accept the proposition that the dosage used here would only be equivalent to 1.5g for a human, the recommended dosage for NR is generally 250mg for a human. So if you were to ingest a dosage 600% higher than recommended, it follows that the kidneys may show some sort of inflammatory pathway activation. This is not shocking but rather common sense. And in general, if the bottle recommends taking 1 pill per day then don't take 6 pills per day and if you do well then your kidneys may need to clean up your mistake. Further, when the authors state "this dosing of NMN is still a more realistic model for the potential for excessive self-dosing, which is enabled by the current availability of many NAD+ precursors as over-the-counter supplements" they are showing bias, suggesting that it is NMN as a compound that is inherently problematic rather than people choosing to massively overdose NMN being problematic. I hope to one day read a legitimate study where more reasonable dosages are used and then we will see what if any harmful effects result from NMN. And I do understand that the bias on this channel is toward rapamycin, the "gold standard for longevity interventions", though I would hope in future to see clear, unbiased reviews of published research. Although I laughed out loud at Matt's reaction to saying Sinclair's name (that was classic), there is nothing to be gained by emulating his behavior even in the limited sense of conducting biased reviews of scientific studies.

I have a theory that our ancestrals always did make fire. So they were constantly heated for hundreds of thousands of years. Therefore, if that heat was bad, natural selection would select species that didn't use fire; But most probably that heat was good in some way and then heat is good for us. other possibility is that has no effect, wich intuitively doesn't make sense.

If you are interested in longevity, ,Bunny, why would you smoke or drink alcohol ??? If you smoke (tobacco) or drink more than two drinks a week- and probably smoking less marijuana (or none) makes sense as well. I mean, are you really wanting to maximize your life span or not. By the way, for myself, I use very little alcohol - usually a mouthful of alcohol held in my mouth while chewing a 325 mg of aspirin. For pain. But I am not totally committed to maximizing my age. I do time restricted eating and try high protein and avoiding food that tastes sweet. But under stress, I do eat ice cream. I am stressed- One of my granddaughters (( mother of three )) was shot breaking two ribs, damaging one lung, and cracking her scapula. She was always going to survive. We now know she will not lose the lung, and they took out the bullet Monday before last, though that wound is still draining. An excuse, I would say, for stress induced comfort eating. I have also lost 40 pounds over the last three years, since adapting this lifestyle. If I can manage to not drink, smoke, or gain weight at my age (( I am 76, )) Buck up, Mr. Bunny, you can decide you don’t need booze and tobacco.

Hello, thanks for the Q&A - I haven't seen much regarding the use (or cessation) of rapamycin during conception, pregnancy and whether or not long term use would affect/influence sperm health or epigenetic factors that may be passed to offspring either through the sperm or egg (or both). Is there cause for concern with no turning back if an individual starts rapamycin prior to starting a family or is there data from transplant patients suggesting that it is not much of an issue?
