Matt, who ran the Kaeberlein lab at the University of Washington from 2006 to 2023, rounded up several former lab members for a chat at the recent 52nd annual meeting of the American Aging Association (AGE) in Madison, Wisconsin. The group included assistant and tenured professors at various institutions around the United States, startup founders, and prospective PhD students with geroscience research interests that span the microbiome, mitochondrial dysfunction, frailty, metabolism, sex differences in aging, and more.
In this episode, Matt and his former lab members pool their collective knowledge to engage in a lively conversation (and sometimes debate) about longevity interventions such as metformin, acarbose, and sauna use. In their discussion, they touch on hot-off-the-press positive results, unpublished data, and planned research from their respective research groups and labs. They also bring up nuances to recent published work, including results from the Interventions Testing Program on various interventions that Matt discussed in a previous podcast episode.

Hi, I'm Matt Kaeberlein. I spent the first few decades of my career doing scientific research into the biology of aging, trying to understand the finer details of how humans age in order to facilitate translational interventions that promote healthspan and improve quality of life. Now I want to take some of that knowledge out of the lab and into the hands of people who can really use it.
On this podcast I talk about all things aging and healthspan, from supplements and nutrition to the latest discoveries in longevity research. My goal is to lift the veil on the geroscience and longevity world and help you apply what we know to your own personal health trajectory. Although I participated in Matt's rapamycin study, I never realized until now what a heavyweight he is in this field. And all your colleagues clearly have a lot of respect for you.

Loved seeing the human side of the Optispan team in this latest podcast. Your personal takes on intermittent fasting were really interesting. Despite the debates around Metformin, my own experience has been amazing: lost a lot of weight, yet got stronger, and as a prediabetic, it’s improved my life in every way. Just goes to show, one size doesn’t fit all, and context really matters

Do you plan on testing rosmarinic acid? "Plant and fungal products that extend lifespan in Caenorhabditis elegans"- Rosmarinic acid, a substance in mint and lemon balm, among other things, increased the life span of the nematodes by 63 percent. Second and third come pomegranate and curcumin. These two substances extended the lifespan by 56 and 55 percent, respectively.

I'm also interested in rosmarinic acid after I saw videos about many centenarians in Acciaroli who used a lot of raw Rosemary to chew on and put it in many dishes. A possible explanation is that it influences Angiogenesis (the growth of blood vessels from the existing vasculature)

@@HvdHaghen I make rosemary tea with dried rosemary, stevia and lemon juice. I also add a small amount of black pepper for the piperine. It may potentially enhance the bioavailability of the rosmarinic acid. The tea tastes great!

I think metformin (immediate release) causes diarrhea much more often than acarbose, and though acarbose does cause flatulence, if you increase it slowly, over time that side effect often stops. And yes, importantly, it does transform your pasta into a salad. lol

It's interesting, as a lay person and long time, avid, science podcast listener, to see how siloed research/ers can be. As an example, the first compound, MB. has been studied in this country by people like Dr. Francisco Gonzalez-Lima, for a long time. I get it though, these folks are doing deep dives into their particular areas of interest and asking questions outside of their areas of expertise can result in speculation and "deer in the headlights' scrambling for safety. That said, I'm really appreciative of this podcast and the rigor with which Matt K. approaches it.

Really appreciate this talk! I'm surprised that so many experts are unsure about so many interventions. Funny that almost everbodies 'weird' longevity intervention is the same. It was probably a joke but the assumption that people are lazy (cold water plunge) excludes a lot of people who do like to work on their longevity and excercise regularly (I assume the type of people that watch this channel).

Seems a bit odd to favor rapamycin but not BCAA restriction as they both affect mTOR1. Would be good to parse. Yes, more work needs to be done. Interesting work from Lamming lab pointing at isoleucine. Downside of HBOT is cost. Hypoxia can be done at no cost.

I think Matt mentioned that high protein (including high BCAA) will likely only activate mTOR transiently especially if you are not having an protein focused energy surplus. I think quantity is the key. Also Dr Sinclair (he is a proponent of inhibiting mTOR as well)mentioned similar opinion on this. After all, we know we need some BCAA. We also know protein, carbs and fats can be interconverted. If you maintain yourself on a slight calorie deficit, the harm from high protein should be very minimal.

I loooove this brilliant format. I wish that you and your panel rated the supplements and methods for daily health as well as the longevity angle I could tell there were some on the fence because they knew the benefits but were measuring for their longevity capabilities. Maybe you could think about doing that. I don’t wanna live long. I just wanna live great. ❤

Lack of exercise, poor dietary habits and all around lack of discipline. I see this exact same thing among many physicians as well.

I am not clear if they think the Longo’s FMD (or even pure fast) is potentially effective for health.

On parabiosis, why not skip straight to exchanging out old plasma with saline as the Conboy lab did. See their 2020 and 2022 papers. Anyone can donate plasma at local blood bank and loose 25% of there old plasma in one go. This needs more study but seems to positively affect many markers including DNA damage. Maybe a good easy therapy for older people along with exercise.

It wasn’t mentioned but should be noted when talking about BCAA restriction, that in the NIH ITP mouse trials, Leucine didn’t extend or more importantly reduce mouse lifespan

Matt believes that Rapamycin is the gold standard at this point. Perhaps your organization could acknowledge Dr. Blagosklonny and Dr Alan Green contribution with Rapamycin.

Hi Matt, I know this is not your primary focus. But can you make a comment about the gene therapy (activating the Yamanaka factors), led by Juan Carlos Izpisua Belmonte and frequently mentioned by David Sinclair. I am fully aware that this type of technology is likely decades away from practices if it will ever be. But let’s also be honest, life extension from simple drugs and lifestyle can only be very modest even if possible. Age reversal will probably rely on something fundamental like in vivo gene editing.

wow.. not even an attempt to think about extending maximum human lifespan/healthspan.. astonishing.. the modern 'longevity science' is a huge nonsensical pile of quasi-scientific nonsense.
