In the comics the gauntlet gives awareness over everything in the universe thought time and makes him faster than everybody. In the MCU it can't even show an incoming danger 5 seconds ahead. The spider sense gives more awareness of the surroundings than the gauntlet.

Even if you brought one to the Marvel universe and it worked, Thanos could classify himself outside of entities, blocks, etc. with the soul stone and thus be unaffected by the commands. Command blocks themselves are also quite easy to remove and take a bit of time to set-up, making them incredibly vulnerable. I don't think they would work against Thanos, although they are a good example of the level of power needed to compete with him since they're able to essentially rewrite the universe.
