Hi Francisco, love your videos, wanted to suggest getting the pc to a boil, before putting the weight on the valve. You should feel steam blowing out. It will get you to pressure in half the time.

@@PlantCellTechnologyOnce sterilised I noted the media cointainers were removed and left to rest on the counter with the lids still on top (unsecured), presumably to allow them to come to room temp to reduce condensation? My question was how long can they safely sit out before securing the lids. I've been concerned of risking contamination and would allow the pressure cooker to come to room temperature (sometimes overnight) prior to opening the PC and sealing the containers. I'm still getting considerable condensation inside the media containers and was wondering if it was due to staying in the PC for an extended period of time.

@@greencleanenvironmentalsol7053 I don't close the containers immediately as they will shrink. I wait around 10-15 minutes before closing the lid. The medium that I make also has PPM, which prevents contamination.

I have only normal domestic cooker without pressure gauge for autoclave..in that how much time should i keep the medium for autoclaving..or how many whistle to be needed...

apart from temperature and time, pressure is also an important factor in autoclaving tools and chemicals. It'd be great for you to purchase a pressure cooker with pressure gauge.

These are autoclavable plastic cups. Regular plastic cups will melt in the pressure cooker because of the high temperature. You can prefer Polypropylene (PP), polypropylene copolymer (PPCO), polymethylpentene (PMP), ETFE, FEP, and PFA made cups to serve the purpose.

Why not just let the steam blow out of the rocker hole for like 10 to 15 min to purge the air out? Why open it back up? Or are you running it two times?

The cooker is only opened after all the pressure is released. In the video, we showed the same.

The All American pressure cooker will get up to pressure, but you have to bleed all the air out. That takes 2-3 minutes, then you let it build pressure again and when it reaches the 15 psi is when start the sterilization process. So, 45 minutes to warm up and build pressure, then 2-3 minutes to bleed the air, and then another 15-20 minutes to build pressure once again, then 40 minutes of sterilization time. and 15 minutes to release the pressure and open the pressure cooker. Total cycle time is close to 2 hours.
