I interpreted the fist pump as just a reaction to how much he was cracking up over the huumba's. The audience stops laughing are quiet for 4:13 - 4:21 and seemed shocked by the kiss. Lol @everyone reading waaaay more into this than it actually is.... The character Stefon has a thing for the other one and had unsuccessfully tried to 'do something' about it before and made off-color comments before. Finally got to give him a kiss (probably scripted as a short smooch that the actor turned into a bigger deal). No closeted anything, nothing between the lines. Just the actors having fun with some sexual tension between their characters. That's why the smile--it was a running gag (reflex) and as another commenter put it, Stefon's actor "won this round". That's all.
