We've heard of the "TATAA" sequences being crucial for RNA transcription. My question is how important is each base of an annotated promoter? Would inserting exogenous DNA into the promoter lead to decreased RNA transcription?

Thanks for your question! You are right that the TATAA sequence is critical for RNA transcription. For the rest of the promoter, it is difficult to say precisely how important each base is and it likely varies depending on the specific promoter. Inserting exogenous DNA into the promoter could certainly decrease transcription efficiency, but to what degree would need to be tested experimentally.
