@@DAAI741 drill a separate hole for either poly fill (have to be careful not to get that wet) or look up hydrophobic syringe filters and google that with mushroom and you should find the size you need and videos on how to drill and attach to canning jars

NOTE: If there is more than 4% sugar to water ratio the liquid can be come toxic to the mycelium/spores. It's better to use less than more if you decide to guesstimate.

Thanks for watching! Store colonizing jars at 70-80 degrees light doesn’t matter but darkness is preferable , once full they will last for 6 months at room temp and can and should be refrigerated to force the mycelium to go dormant for longer than a year.

@@jeff042854 Liquid culture is just spores that have been germinated by introduction to sterile nutrient media. Spore syringes are made with only water to prevent them from germinating, they can be store for years before use.

Some of the bags look like they're about done and some look like they're about halfway done. How do I know when to move the cake to the mono tub? Should I do each one as it's ready, or do I wait till they are all done? I'm going to be watching your videos because this is the first one that I've seen, but if you can answer my question I would be very much appreciative for it. They are already talking to me, I'm excited!

Its best to use liquid culture as spores take up to a week or so to turn into mycelium and spores are also usually contaminated so its best to do a brf tek with the spores, get agar, take a piece of clean mycelium from either the middle of a cake or the middle of a mushroom or something and place it into the agar, once it turns to mycelium you can use that to create your own liquid culture, you have a much greater risk of contam and contam is very hard to see usually in liquid

1. It is not definitive, but most aggressive species will completely fill the liquid culture jar with mycelium. Once the jar is full of thick mycelium it should be stored in a cold room or in refrigerator. Some species like lions mane and shiitake will usually only grow half way in the liquid culture jar and are not as aggressive. 2. Storage at room temp is fine for up to 6 months, refrigerated or cold room shelf life is over 12 months. It’s hard to tell when it has gone bad, but a way to test it is to put a drop on agar plates. If it grows it is still viable. If the culture is very old it will be noticeably slower or yields will be smaller and smaller over time. 3. Honey is way better than karo in my experience. Karo produces more thin, weaker mycelium and honey produces very thick, aggressive mycelium. It is good to switch between honey and LME once every 1-2 years to change up food source. Got a stir machine for less than $30. Its worth it. Make sure to put the stir bar in before sterilizing, but sanitize the bars first. I think they were the source of contam the first time.

Hi my friend I was just watching you on KZbin and I will say I was very impressed by your knowledge and willing to share with us. My son has been growing mushrooms for me to eat because of the research he has been doing about the health benefits of a few different types.... I have a question. We are in the process of making the jar myzilyume. We are in Utah so it's extremely cold. For two to three months his been trying to get some to grow. My question is how long will the myzilyume last in the jar before it's not good to grow anything. We want to be ready for when the weather changes and it's warm enough to grow them. We were trying for a long time and finally I told him to go ahead and keep the room really warm and let's see what happens. Two weeks later we were enjoying some kind mushrooms lol thank you

I will say that my current jar of liquid culture was made with no magnetic stirrer and no marble and the entire thing is evenly cloudy. I just spun the jar with my wrist at least 1x per day for the past 12 days.

You would inoculate grain spawn with the syringe and once the grain spawn is colonized, mix with substrate. Yes each bag will be identical. I have other videos that show the whole process.

Yes I can do that. I’ll start working on it ASAP. In the mean time, quick sneak preview would be to tilt the jar of colonized liquid culture and inject empty syringe and suck up the culture that way. The only problem with that is, if your mycelium isn’t very dense and full in the jar it will be hard to reach. If that happens, try using LME recipe l (I will be making a vid on that soon too). Or you can super sanitize your work area and also use a UV-C light to sterilize everything and then simply open the jar lid and suck the culture up and close the lid right after. Hope this helps.

@@SporenSprout yes this helps but I’m also trying to figure out what syringes you purchase for this exactly like you order the syringes, extract the liquid culture and the replace the syringe with a stopper under a flow hood and then send a sterile packaged syringe to a buyer along with the stoppered liquid culture correct? Also when is your LC considered colonized enough to start extracting it? I have a weeks worth of lions mane LC that I stir everyday. When can I start extracting it? Should I wait a few more weeks of mycelium accumulation? And also how do I prevent it from forming hyphal knots?

@@HoneyandMyco order empty sterile 10 ml syringes from Medvet international (they come with needle attached) and a separate order of 18 gauge sterile needles, and syringe caps. Open the syringe package near flow hood or in SAB and suck the culture up, take needle off and replace with syringe cap. Now the syringe is ready to be labeled and packaged along with one sterile needle. That is all the friend or customer would need. One capped syringe of culture along with one sterile packaged needle. There is no set maturity to the Liquid Cultures. As soon as there is enough to fill the syringe with a decent amount it is good to go. Usually takes me about 1-2 weeks for adequate growth . Use a magnetic stirrer to insure they mycelium can be broken up and distributed for faster growth and also fresh air sucked in my vortex it creates.

Yes you can put a few ml in more LC media or you can put a few drops on a bunch of agar plates to multiply that way. There is no definitive answer to your question, it can be done many times until it stops performing well from old age, some species degrade quicker like cordyceps.

If you send spores to liquid culture, the liquid culture will have many different genetics so your fruit bodies will be not very consistent. When you send spores to agar you have the ability to choose the best growing sector of mycelium and transfer to a new plate. Then you can use a wedge of the new plate to send to liquid culture. Then your liquid cultures genetics will be more consistent. Not a big deal either way though. After replicating liquid culture many times you will have to start back again from spores to keep the culture fresh and strong.

I've got a small cloud forming in jar....now to keep it going, how warm should the room be for it to thrive? It's cold up here in Oregon, should I get a heat pad?

Nice!! You should stir the cloud every few days so that it breaks apart into tiny pieces. I use a space heater in my room and set it at 70-75 F

In a perfect world, you would have a dedicated mini fridge just for culture but to answer your question, I don’t use a plastic bag and I don’t know that anyone else does. If the liquid culture is super thick and full like it’s supposed to be, there is less room for contam also.

You can watch my other videos for information about how to use the liquid cultures. A syringe is used to extract the liquid culture after 2-4 weeks of growing in the liquid. Can be stored for years at room temp or refrigeration

You can use spores but you will have many different strains growing. Also spores are not entirely clean so you may have to test the final product on agar to see if it’s clean.

Great video! I don't have a pressure cooker or a magnetic stirrer. I know they're widely used in mycology but I was wondering if there's any way to sterilize and stir without investing in those items? For example a make shift steamer using everyday household pans (instead of the pressure cooker). And simply physically shaking the jars (instead of using the magnetic stirrer). Any broke boi possibilities there?

You can steam the jars for 2 hours in a pot of water with a lid. You cannot shake the jars if you use a micropore filter because the liquid will touch the filter or even leak from it. You can use a marble and swish it around the bottom to help break up the mycelium instead of shaking or using a magnetic bar.

What is the difference between this method and your another method of a separate injection port plus air filter hole and pressure cooking with 15 psi for 30 minutes ? Also have a question about the regular 2 piece mason jar lid that rusted the filter hole overnight after i left the honey water in the jar for overnight cool off. i am using your liquid culture syringes which i bought from etsy. Do you think i will have to redo everything over again with plastic lids or it will be still okay.

The difference is this video is I didnt use rtv silicone injection port. The filter hole was not rusted, you probably splashed the honey water and it came in contact with the filter. The filter should not get wet. Make sure water level is half way in jar, make sure not to over cook too hot, make sure not to splash. If the filter gets wet, dry it off with laminar flow air.

If you invert your lid and put it in a hot oven it will melt stuck to the ring. That makes it easy to remove when you inject spore solution.

Using 2 ml per inoculation, you can inoculate 5 3 LB bags of grain which you can grow around 1 pound of dry mushrooms from. You can attempt “grain to grain transfers” to spread the mycelium to even more bags of grain and exponentially increase output.

Months. It’s actually good to let it sit for a few days before inoculation just to make sure no other growth appears. Btw you should use 1 Tablespoon of honey instead of karo. It’s much better.

@@7H3M4DH8R Yes this is very common due to over cooking. You should start over, It has to be done very precise. Use very warm but not boiling water and dissolve 1 tablespoon. Pressure cook 30 mins and not a second longer. Try not to go over 15 psi as well.

I have two quart jars, one with blue oyster and one with lions mane. The blue oyster mycelium is growing every day and after two weeks, has filled up nearly half of the liquid medium. The lions mane, however, grew a little in the beginning, but seems to have stalled out in the last week or so. Is this an issue with the mycelium or could this just be a species specific issue?

No I used a liquid culture syringe. Liquid culture is basically germinated spores. It is mycelium. You can use a spore syringe the same way as you would a liquid culture syringe but it is considered multispore inoculation and will result in many genetic differences.
