Hi, thanks for your excellent tutorials! Very nice to watch and high instructive. With regard to sterilizing in the pressure cooker, what is there against putting a second layer of jars on top of the first layer, again with a spacer to stop jars from touching. Thanks, michael

Hi everyone. I'm seeking a pressure cooker for sterilization in my small lab at home but I can't find one with pressure meter (which is necessary to see when it gets 15 psi and 121 degree C). I heard that usual cookers in market gets around 15 psi when the pressure regulator (or rocker, the little thing we put on the vent pipe on the lid) starts to rock. Is it true? Can I actually use a pressure cooker without pressure meter for sterilization at 15 psi and 121 degree C? Thank you all very much 🙏

Loved the video! Could using this method to sterilize the media possibly cause evaporation of the chemicals in the media? Im just worried about doing this in my home because I don't want any of the PGRs in the air.

Where did you get the poly lids that are autoclave-able that fit wide mouth mason jars?

Hi Laur, very nice to watch your informative vedios. When we use plant growth regulators in media how long can we store media after pouring in sealed containers while these are mostly thermoliabe. For example if I am using rooting media and I add 2 mg/L IAA stock solution, after solidification to inoculation, will they not lose it's efficiency?

I think the media should be good to use for at least a few months as long as you seal the containers tightly to prevent contamination and evaporation. The efficacy of plant growth regulators can be affected by a number of factors, including the storage conditions, the age of the media, and the type and concentration of the growth regulator used, so probably ideal to use it within a few weeks of making it

I found that, unless I let the cooker internal temp to go to baseline, my caps and jars are slightly offsized and difficult to thread. Having to fumble with them introduces the potential for advantageous pathogens.

@@chugginbeers Sorry, when I said 'advantageous' I meant taking advantage of the open top. However, there are beneficial microbes that live in symbiosis with a number of carnivorous species. One of the best known is the relationship between heliamphora and trichoderma. The presence of these in the cells of the hosts can make sterilizing an explant very difficult.

Only 2? Poser! I have 8. hahahah Grab some stainless steel nuts, washers and bolts to make a trivet out of the canning rack. Venting tip: Let it build to 10psi then, remove the rocker until psi drops to near zero.

@@plantsinjars Three are benchtop units with their own heating elements. One, I'll probably give and teach how to use, to my daughter. She wants to do what you're doing. So, sterilizing and agar work will come from me. I grow different mushrooms.

I use an instapot successfully with polypropylene containers or bell canning jars with polypropylene lids. Only downside is the lids are opaque. Zero contamination even stored for a year if they are unopened. Also, I have an All American pressure cooker. It doesn’t have a rubber gasket and is basically is bulletproof as long as the sealing surfaces don’t get dinged. I’ve owned several other PCS like Presto and Kuhn and hate the gaskets that always eventually fail at an inopportune moment.
