A short film showing how to use a Gilson Micropipette. The film was made to support the teaching of undergraduate Biological Science students at the University of Leicester.
The film was produced by GENIE, based within the Genetics Department at the University of Leicester. Further information and materials can be found on the GENIE web site www.le.ac.uk/genie and the GENIE VGEC web site www.le.ac.uk/vgec
The film was directed by Jon Shears of ITS-Multimedia Services and the film was produced by Raymond Dalgleish, Nicola Suter-Giorgini and Cas Kramer.

P2 range: 0.2-2ul P20 range: 2-20ul P200 range: 20-200ul P1000 range: 100-1000ul (in this case u is the symbol for micro, or 10^-6)

You've covered the possible ways the pipette can be contaminated or damaged,but you've failed to follow up on what to do about it. I started working in an analytical lab over 20 years ago and they were having trouble passing the unknown quantitative metals tests each year required by the EPA. My first year on these analysis's I was testing the output of my eppendorf pipette on an analytical balance with reagent grade water to insure in was delivering the proper volume before making my standards. Another analyst asked me what I was doing , I told them and they then said." well I worked in the medical field for over 10 years and we never tested our pipettes". All I could think was no wonder so many people die in hospitals if this is normal SOP in the medical field. If the pipette is found to be not delivering the set volume it must be taken apart and cleaned or repaired !!!! After this all our measuring equipment was checked closely for accuracy. We never failed our yearly test again .

The first thing I did on receiving a new pipette was to test it. a) for accuracy, b) for repeatability and c) to check my technique. That analyst wasted 10 years.

not bad for an instructional video. these science instructions are usually 20 years old and out of date. this video was really well done--with camera angles, explanation, and reasonings

If we have to use the second stop to eject the volume into the receiver tube there is going to be bubbles with that which is going to give you inaccurate results when say you are measuring the OD of serum samples. And if we don't use the second stop there would still be liquid in the pipet tips. What do we have to do?

Hi may I ask does the Pipette need to be vertical at all times even when dispensing liquid and Does the pipette tip need to be against the wall of the receiving tube during dispensing ? Thank you

I've been told to repeatedly wet the inside of the pipette tip by carefully pipetting up and down to the first stop 2-3 times with the liquid to get the accurate volume, is this not necessary?

Nice explanation. However I am confused at 2:44 so if I have to 152 uL using P1000, then what should be the number on pipette?

You couldn't set the P1000 to 152 µL, it would only let you set the µL in sets of five, so you could set 150 and 155, 160 and so on.
