Yea... you can tell during rehearsals John said that line differently. Then when it came time to go Live, he switched it up. I mean his character's name is SHY.... and he just overly confident "Hello gentlemen. How are you" threw off Bill a little.

John Mulaney created the Stefon character with Bill Hader. John always loved it whenever Bill would break character during the skits, so he used to try to throw him off by replacing some of his lines on the cue cards at the last minute so he'd be reading a joke for the first time. Since John no longer works there and Bill invited him to come do this cameo character, John decided to just say something ridiculous to throw him off again.

Seth used to be the news correspondent on SNL's "Weekend Update" and he'd continuously invite Stefon, where they would both shamelessly flirt. Stefon attempts to do "A Closer Look" here and is asked why he's doing Seth's bit. Stefon insinuates that they roleplay (sexually). So instead of Seth being on top and Stefon being under the desk, the roles are reversed. It's meant to be a tongue-in-cheek joke at Seth's expense. If you're confused about what one of them would be doing under the desk, let's just say that one would be giving an "oral" presentation. It's a dirty joke.

A part of me was thinking about Mulaney, and how he was the head writer for Stefon. And then he SHOWS UP! It makes me so proud that he’s in front of the camera for SNL considering all of his success since being behind the scenes. I’ve haven’t even seen John and Bill in the same camera shot before, only great stories about their friendship in interviews. I missed this so much.

They aren't his jokes - when he's doing Stefon he's reading them for the first time which is why he breaks up most of the time. They are primarily written by John Mulaney.

I love that he just gave up at some point and was just enjoying the moment instead of fighting for staying in character when he's simply not up to it. I think it's brave and it definitely works. There's a certain magic in letting go and just being in moments like these.

Considering how Bill Hader has spoken out about his issues with anxiety and stage fright, it's also just a huge victory for Bill Hader, personally. I can't imagine how liberating it must have felt to finally go out onstage and just not care about messing up or about what the audience thinks.
